A novel pH-stable, endoglucanase (JqCel5A) isolated from a salt-lake microorganism, Jonesia quinghaiensis Electron. J. Biotechnol.

Background: Endoglucanase, one of three type cellulases, can randomly cleave internal p-1,4-linkages in cellulose polymers. Thus, it could be applied in agricultural and industrial processes. Results: A novel endoglucanase gene (JqCel5A) was cloned from Jonesia quinghaiensis and functionally expressed in Escherichia coli Rosetta (DE3). It contained 1722 bp and encoded a 573-residue polypeptide consisting of a catalytic domain of glycoside hydrolase family 5 (GH5) and a type 2 carbohydrate-binding module (CBM2), together with a predicted molecular mass of 61.79 kD. The University of Washington PBL model is a nonlinear similarity model (with organized large eddies in an equilibrium mean flow) suitable for km-scale to General Circulation Model or Climate numerical models. The theory and verification for this solution are now well established. Researchers who work with winds in the boundary layer with scales from point to 20-km regimes must be aware of this horizontal inhomogenuity in the PBL flow to avoid significant errors. Since the OLE contribute advectively to the mean flow there can be 10-20% (or more) increase in surface winds and fluxes over K-theory models. Here we introduce a new approach for the screening of DNA binding proteins, using a phage library based on a phage display technique. In principal, a complementary DNA (cDNA) library based on the recombinant bacteriophage T7 expressing target proteins on its capsid (phage display) is constructed. These phage particles are hybridized with a biotinylated target DNA fragment which is immobilized on the surface of streptavidin paramagnetic particle (SA-PMP). The phage particles are released from the target DNA fragment by a nuclease treatment and the recovered phages are used to the next round of hybridization. To evaluate the mandibular angle and to analyze the relationship of the angle and height & breadth of the ramus of the mandible to the gender, so as to study its role in the anthropological diagnosis. The angle, height and breadth of the ramus of adult dry human mandibles of both sexes were measured using a goniometer. The values obtained were analyzed statistically. The present study showed a statistically significant difference in the mandibular angle as well as height of the ramus between both the sexes. Diabetic nephropathy (DN) is one of the major complications of type 2 diabetes and is associated with coronary disease. Nephrin, a protein mainly expressed in glomeruli, is decreased in DN and other kidney diseases. Since insulin levels are misregulated in type 2 diabetes, a possible connection between DN and its decreased nephrin expression could be the presence of regulatory elements responsive to insulin in the nephrin gene (NPHS1) promoter region. Cytochrome c oxidase, the terminal enzyme of the respiratory chains of mitochondria and aerobic bacteria, catalyzes electron transfer from cytochrome c to molecular oxygen. The enzyme belongs to the haem-copper-containing oxidases superfamily. A recombinant plasmid carrying a 2.0 kb insert from a Burkholderia pseudomallei genomic library was subjected to automated DNA sequencing utilizing a primer walking strategy. Complementary DNA (cDNA) is valuable for investigating protein structure and function in the research of life science, but it is difficult to obtain by traditional reverse transcription. In this study, we employed a novel strategy to clone the human leukemia inhibitory factor (hLIF) gene cDNA from genomic DNA directly isolated from the mucous membrane of mouth. The hLIF sequence can be acquired within a few hours by means of amplification of each exon and splicing using overlap-PCR. Thus, the new approach developed in this study is simple, time- and cost-effective, and it is not limited to particular gene expression levels of each tissue.

A Pyranine based Procedure for Evaluation of the Total Antioxidant Potential (TRAP) of Polyphenols: A Comparison with closely related Methodologies Biol. Res.

A novel procedure for the evaluation of total reactive antioxidant potentials (TRAP) is described. The method is based on the measurement of the bleaching of pyranine by peroxyl radicals. The addition of the antioxidants produces a clear induction time whose magnitude is directly related to the antioxidant concentration. A comparison of the values obtained with those reported employing closely related methodologies shows that the results are significantly affected by the substrate employed to monitor the steady state free radical concentration. A Quantitative Assessment of the Morphological Characteristics of BeWo Cells as an in vitro Model of Human Trophoblast Cells International Journal of Morphology

In order to study the detailed morphology of trophoblast cells during human implantation, BeWo cells were cultured as spheroids in suspension culture. These cultures were then processed for light and electron microscopical examination. The present study showed that the BeWo spheroids consist of two cell types which are cytotrophoblast-like and syncytiotrophoblast-like. The cells with larger nuclear diameter made up only about 1% of the cell population and appear to be those of syncytiotrophoblast. Therefore the predominant cell type of the BeWo spheroids appeared to be relatively undifferentiated and cytotrophoblast-like. A radiographic method to estimate lung volume and its use in small mammals Biol. Res.

In this paper we develop a method to estimate lung volume using chest x-rays of small mammals. We applied this method to assess the lung volume of several rodents. We showed that a good estimator of the lung volume is: V*L = 0.496 · V RX ≈ 1/2·V RX , where V RX is a measurement obtained from the x-ray that represents the volume of a rectangular box containing the lungs and mediastinum organs. The proposed formula may be interpreted as the volume of an ellipsoid formed by both lungs joined at their bases. When that relationship was used to estimate lung volume, values similar to those expected from allometric relationship were found in four rodents.
